Do you see this spot when candling, or just from looking at its surface? If not try candling the egg and see if you can see the embryo. At this point you should be able to make out its head, eyes and body, maybe even some movement through the shell. Eggs can still swell up regardless if the embryo is present or dead. If you do check be very careful not to turn the egg, if the embryo is alive it can easily drown.
