I agree with Iguana Joe in that the blood sounds a bit alarming but the act is pretty natural. How much blood was there and were there certain patterns you can decipher? I'm asking if maybe they were small amounts of blood from his gums/mouth from biting the mit too hard. This would happen to my last iguana who tried to eat everything in sight. I found blood on newspapers where he would take chunks out of....teeth marks and blood. It would hurt his mouth. I'm not sure if an oven mit could do this since it is softer, but I suppose it is possible. If it is a good concentration of blood (like a large stain) then I would say it probably was from his vent. I suppose excessive humping could have caused it. I've seen old blood on the end of seminal plugs. Just keep an eye on it. If the bleeding doesn't stop, obviously take him to the vet.
