Without seeing the enclosure I'm taking shots in the dark but as far as hardware, I'd say 1" or 1 1/4" screws with finish washers would do the trick. Any drill bit should work. If you're going out to buy one specifically for this job get a brad point bit. It will grab easier and won't "walk" on the plexi. Without sealing the glass with a bead of silicone, though, it may not be as efficient as you'd like. Of course, if you DO use silicone, it could turn into a permanent installation. Quick thought, what about adhesive backed magnetic strip? Assuming that the adhesive sticks sufficiently to both plexi and wood you'd have a fairly good seal and the panels would still be removable.
