if its a he, he should have develop large jowls by now, not as big as the male pic in the post below, but quiet noteceable.
Also, bluges of his hemipenes (igs have 2 penises) around the vent area should be visible.
U can have him/her probed by a professional, or X-Rayed. There are methods of "massaging" the vent to make "it" come out, but it is not reliable, and may not always work. Also, if its a he, it should start to act aggressive during mating/breeding season, and show more color at those times. Look for seminal plugs around the enclosure (dry semen).
It's not easy, ut at 4 years of age, it is much easier! Iguanas can be usually seed with some success at 2 years of age.
